To directly quote the official press release (http://e3.nintendo.com/pressrelease/),

After going dark six years ago, 2010 will see a new dawn. Golden Sun DS updates the popular portable Golden Sun role-playing series with an impressive graphical style. This installment follows the story of the previous heroes’ descendants and immerses players into the magic of its adventure by pushing the boundaries of intuitive touch-screen controls. Golden Sun DS is playable on the Nintendo DS and Nintendo DSi systems. Descendants of the original cast, meaning Isaac is not the protagonist. In all likelihood, the original group, with one possible exception, is going to be dead. Quite dead. Nothing to help determine anything else about the plot.

Gameplay video (http://www.youtube.com/watch?v=Lm3-RRgXtnI&fmt=18) shows Atlanta, Ramses, what may or may not be Nereid or a new summon completely, and Eclipse. It also shows potentially aimless wandering. No psynergy is shown.

Unless I've missed some materials, which is perfectly possible, there's not much else to know. Descendants of the original cast, includes a few of the old summons and at least a couple old enemies, house in the middle of nowhere.

Still, nice to know it's probably going to actually have a third game released.
